Study Design

This was an open-label one-sequence crossover study involving n=12 health adult subjects. Each received a single dose of valsartan 80mg alone or in combination with glecaprevir/pibrentasvir 300mg/120mg QD. Intensive blood sampling for determination of metabolite and glecaprevir/pibrentasvir concentrations was done. The effects of glecaprevir/pibrentasvir on the PK of valsartan and vice versa was analyzed.

Study Results

Exposures were slightly higher for valsartan with an increase in Cmax 36% and AUC 31% when administered with glecaprevir/pibrentasvir. Exposures of glecaprevir/pibrentasvir were similar regardless of if valsartan was onboard or not. This combination was generally well tolerated, with no serious adverse events being reported.

Study Conclusions

Per the authors, clinically significant PK interactions between glecaprevir/pibrentasvir and valsartan are not anticipated. No dose adjustment is required when glecaprevir/pibrentasvir and valsartan are coadministered.
