Summary

The authors noted that there was a lower mean creatinine clearance in the kanamycin group versus the control group (91mL/min vs. 112mL/min, respectively), and this likely accounted for the observed changes in the AUC. They concluded there was not a clinically relevant drug interaction between kanamycin and bedaquiline.

Study Design

Patients with multidrug resistant tuberculosis (MDR TB) (n=16) received 14 days of bedaquiline 400mg daily or placebo followed by the administration of 1,000mg (normalized daily dose) of kanamycin. Pharmacokinetic parameters were taken over 1 dosing interval.

Study Results

The ratio of the AUC in the study vs control group for kanamycin was 1.51 (90% CI 1.15 to 1.98).
