Summary

A single dose of etravirine (ETV) 100mg was given to healthy subjecs during the first period of a 3 period one-way cross-over study; period 2 was an 11 day course of ranitidine 150mg twice daily, with a single dose of ETV 100mg on day 8; period 3 was an 11 day course of omeprazole 40mg once daily, with a single dose of ETV 100mg on day 8. The AUC and Cmax of ETV when co-administered with ranitidine were 86% and 94% respectively compared to ETV alone. The AUC and Cmax of ETV in the presence of omeprazole were 141% and 117% compared to ETV alone. The increased exposure of ETV when co-administered with omeprazole was not found to be clinically significant. No dose adjustments were recommended when ETV is given with H2-antagonists or proton pump inhibitors.[159]
